Abstract
The present invention provides a bioluminescence resonance energy transfer (BRET) detection system characterised by a broad spectral resolution between donor and acceptor emission wavelengths. The broad spectral resolution between the emission wavelength of the bioluminescent donor protein and the fluorescent acceptor molecule results in an increased signal-to-base ratio and dynamic range in comparison with a basic BRET system.
Claims34
A bioluminescence resonance energy transfer (BRET) system comprising:
- (a) a bioluminescent donor protein (BDP) attached to a first molecule or modulator;
- (b) a fluorescent acceptor molecule (FAM) attached to a second molecule or modulator, wherein said FAM can accept the energy from the BDP when they are associated, in the presence of the appropriate substrate;
- and
- wherein a physical change in the modulator(s) influences the energy transfer efficiency between the BDP and the FAM and wherein said system has a broad spectral resolution of at least 80 nm between the peaks of the BDP and FAM emission spectra.
