Abstract
Composition and methods for enhancing oligonucleotide-directed nucleic acid sequence alteration in vivo, ex vivo and in vitro are presented. These methods and compositions involve cells and cell-free extracts with altered levels or activities of a protein from the RAD52 epistasis group, the mismatch repair group and/or the excision repair group.
Claims18
A method of enhancing oligonucleotide-mediated nucleic acid sequence alteration efficiency comprising using a cell or a cell-free extract with increased levels or activity of at least one protein selected from the group consisting of a homolog, ortholog or paralog of RAD10, RAD51, RAD52, RAD54, RAD55, MRE11, PMS1 or XRS2.
A kit comprising an oligonucleotide capable of directing nucleic acid sequence alteration and at least one purified protein which is selected from the group consisting of a homolog, ortholog or paralog of RAD10.
A kit comprising at least one cell or at least one cell-free extract wherein the cell or cell-free extract comprises increased protein levels or activity of at least one protein selected from the group consisting of a homolog, ortholog or paralog of RAD10, RAD51, RAD52, RAD54, RAD55, MRE11, PMS1 or XRS2.
A kit comprising a collection of at least two different cells wherein each cell has increased protein levels or activity of a different protein selected from the group consisting of a homolog, ortholog or paralog of RAD10, RAD51, RAD52, RAD54, RAD55, MRE11, PMS1 or XRS2.
