Abstract
Various antibodies with different binding characteristics for MBL A or MBL B bound to mannan or MBL A or MBL B bound to an antibody against MBL are provided. Also provided are methods for selecting antibodies against MBL with different binding characteristics and methods and kits for using these antibodies to measure MBL capable of binding to mannan or oligomerized MBL and abnormal, poorly oligomerized MBL in a sample. The methods and kits are useful in diagnosing increased susceptibility to and exacerbation of infections and autoimmune diseases.
Claims14
A method for selecting monoclonal antibodies raised against MBL comprising assessing an antibody raised against MBL for its ability to bind or not to bind to MBL A bound to mannan or MBL B bound to mannan.
A method for selecting monoclonal antibodies raised against MBL comprising assessing an antibody raised against MBL for its ability to bind or not to bind to MBL A bound to said antibody or a different monoclonal antibody against MBL or to MBL B bound to said antibody or a different monoclonal antibody against MBL.
An antibody raised against MBL, said antibody being selected from the group consisting of an antibody which:
- (a) binds or does not bind to MBL A bound to mannan;
- (b) binds or does not bind to MBL B bound to mannan;
- (c) binds or does not bind to MBL A bound to said antibody or a different monoclonal antibody raised against MBL;
- and
- (d) binds or does not bind to MBL B bound to said antibody or a different monoclonal antibody raised against MBL, with the proviso that the antibody is not HYB 131-01.
A method for diagnosing increased susceptibility to and exacerbation of infections and autoimmune diseases comprising measuring MBL that is capable of binding to mannan in human plasma or serum samples.
A method for diagnosing increased susceptibility to and exacerbation of infections and autoimmune diseases comprising measuring both normal oligomerized MBL and abnormal, poorly oligomerized MBL in human plasma or serum samples.
