Abstract
The present invention provides targeted dendrimeric reporter molecules that provide amplified signals at electrochemical sensors, and methods of synthesis and methods of use thereof. The reporter molecules comprise a targeting moiety and at least one dendritic signal amplifier; the dendritic amplifier comprises a plurality of dendritic branches and a plurality of indicator moieties, typically pendant therefrom. The targeting moiety serves to concentrate the dendritic amplifier at a selected target, where the plural indicator provide increased signals.
Claims39
A targeted dendrimeric reporter, comprising:
- a targeting moiety;
- and
- at least one dendritic signal amplifier,
- wherein each of said at least one dendritic signal amplifiers is linked to said targeting moiety.
A targeted dendrimeric reporter, comprising:
- targeting means;
- and
- dendritic signaling means bonded thereto.
A method of detecting the presence of a chosen analyte in a sample, the method comprising:
- contacting a sample to be queried for the presence of a chosen analyte with a composition that comprises a targeted dendrimeric reporter, said dendrimeric reporter comprising a targeting moiety and at least one dendritic signal amplifier;
- and then
- electrochemically detecting the indicator moieties of the dendrimeric reporter molecules that are targeted to the chosen analyte.
In a method of electrochemical detection of an analyte, the improvement comprising:
- contacting the electrochemical sensor, in the presence of analyte, with a targeted dendrimeric reporter, said dendrimeric reporter comprising a targeting moiety and at least one dendritic signal amplifier.
A method of making a targeted dendrimeric reporter, the method comprising:
- linking a targeting moiety to a dendritic signal amplifier through a coordinating metal center.
A kit for electrochemical detection of an analyte, comprising:
- at least one composition comprising a targeted dendrimeric reporter;
- and
- indicia that identify at least one parameter required to detect the indicator moieties of said dendrimeric reporter.
