Abstract
The invention relates to isolated CGTases und isolated nucleic acid sequences encoding the CGTases. The invention also relates to nucleic acid constructs, vectors, and host cells comprising the nucleic acid sequences as well as methods for producing and using the CGTases, in particular to cleaning and detergent composition comprising such CGTases.
Claims34
An isolated cyclomaltodextrin glucanotransferase (CGTase), selected from the group consisting of:
- (a) a CGTase having an amino acid sequence which has at least 65% 80% identity with the amino acid sequence shown as amino acids 1 to 679 of SEQ ID NO:2;
- (b) a CGTase which is encoded by a nucleic acid sequence which hybridizes under low high stringency conditions with
- (i) a complementary strand of the nucleic acid sequence shown as nucleotides 103 to 2139 of SEQ ID NO:1, or
- (ii) a subsequence of (i) of at least 100 nucleotides.
- (c) a protein derived from the mature peptide of SEQ ID NO:1 by substitution, deletion or addition of one or several amino acids and having CGTase activity;
- and
- (d) a CGTase which has optimum activity in the pH range of from 8.5 to 9.5.
An isolated cyclomaltodextrin glucanotransferase (CGTase), selected from the group consisting of:
- (a) a CGTase having an amino acid sequence which has at least 80% identity with the amino acid sequence shown as amino acids 1 to 679 of SEQ ID NO:2;
- (b) a CGTase which is encoded by a nucleic acid sequence which hybridizes under high stringency conditions with
- (i) a complementary strand of the nucleic acid sequence shown as nucleotides 103 to 2139 of SEQ ID NO:1, or
- (iii) a subsequence of (i) of at least 100 nucleotides.
- (c) a protein derived from the mature peptide of SEQ ID NO:1 by substitution, deletion or addition of one or several amino acids and having CGTase activity;
- and
An isolated nucleic acid sequence encoding a CGTase, selected from the group consisting of:
- (a) a nucleic acid sequence having at least 65%80% identity with the nucleic acid sequence shown as nucleotides 103 to 2139 of SEQ ID NO:1;
- and
- (b) a nucleic acid sequence which hybridizes under low high stringency conditions with
- (i) a complementary strand of the nucleic acid sequence shown as nucleotides 103 to 2139 of SEQ ID NO:1, or
- (ii) a subsequence of (i) of at least 100 nucleotides;
- and
- (c)a nucleic acid sequence derived from the mature peptide encoding nucleic acid sequence shown as nucleotides 103 to 2139 of SEQ ID NO:1 by substitution, deletion or addition of one or several amino acids and encoding a protein having CGTase activity.
A nucleic acid construct comprising the nucleic acid sequence of any of claims 9-127-10 operably linked to one or more control sequences capable of directing the expression of the CGTase in a suitable expression host.
Use according to claim 2423, wherein the cyclodextrin is selected from the group consisting of delta-cyclodextrins, epsilon-cyclodextrins, zeta-cyclodextrins and mixtures thereof.
Use according to any of claims 30-3128-29, wherein cyclodextrins are generated in situ.
