Abstract
The invention relates to a vaccine comprising a pharmaceutically acceptable carrier or diluent and a bacterium, wherein the bacterium contains (i) a DNA sequence encoding a site-specific recombinase, and (ii) a plasmid comprising a recognition element for the recombinase and a DNA sequence encoding a heterologous polypeptide. The recombinase stabilises inheritance of the plasmid and thereby prevents the bacterium from losing the ability to express the heterologous polypeptide.
Claims30
A vaccine comprising a pharmaceutically acceptable carrier or diluent and a bacterium, wherein the bacterium contains:
- (i) a DNA sequence encoding a site-specific recombinase;
- and
- (ii) a plasmid comprising a recognition element for the recombinase and a DNA sequence encoding a heterologous polypeptide.
A vaccine according to any one of the preceding claims, wherein the site-specific recombinase is a member of the resolvase family of site-specific recombinases.
A vaccine according to any one of the preceding claims, wherein there. is a transcriptional termination sequence between the recognition element and the promoter of the gene encoding the heterologous polypeptide.
A vaccine according to any one of the preceding claims, wherein expression of the polypeptide is driven by the nirB, pag C, ssaH or htrA promoter.
A vaccine according to any one of the preceding claims, wherein the bacterium is from the genus Escherichia, Salmonella, Shigella, Vibrio, Neisseria, Bordetella, Listeria or Mycobacterium.
A vaccine according to any one of the preceding claims, wherein the bacterium is an attenuated strain of a pathogenic bacterium.
A bacterium as defined in any one of the preceding claims.
