Abstract
Purified BMP-11 proteins and processes for producing them are disclosed. Recombinant DNA molecules encoding the BMP-11 proteins are also disclosed. The proteins may be useful in regulating follicle stimulating hormone, such as for contraception. In addition, the proteins may be useful for the induction of bone, cartilage and/or other connective tissue.
Claims27
An isolated DNA sequence encoding a BMP-11 protein.
An isolated DNA molecule having a sequence encoding an BMP-11 protein which exhibits BMP-11 activity, said DNA molecule comprising the DNA sequence selected from the group consisting of:
- (a) nucleotide #375 to #704 of SEQ ID NO:1;
- (b) nucleotide #760 to #1086 of SEQ ID NO:10;
- and
- (c) naturally occurring allelic sequences and equivalent degenerative codon sequences of (a) or (b).
An isolated DNA molecule encoding a BMP-11 protein, said DNA molecule comprising nucleotide #375 to #704 of SEQ ID NO:1 or nucleotides #760 to #1086 of SEQ ID NO:10.
A method for producing a purified BMP-11, protein said method comprising the steps of:
- (a) culturing a host cell transformed with a DNA molecule comprising the nucleotide sequence encoding a BMP-11 protein;
- and
- (b) recovering and purifying said BMP-11 protein from the culture medium.
A purified BMP-11 polypeptide comprising the amino acid sequence from amino acid #1 to #109 as set forth in SEQ ID NO:

A purified BMP-11 protein produced by the steps of
- (a) culturing a cell transformed with a DNA molecule comprising the nucleotide sequence from nucleotide #375 to #704 as shown in SEQ ID NO:

- and
- (b) recovering and purifying from said culture medium a protein comprising the amino acid sequence from amino acid #1 to amino acid #109 of SEQ ID NO:

A purified BMP-11 protein produced by the steps of
- (a) culturing a cell transformed with a DNA molecule comprising the nucleotide sequence from nucleotide #760 to #1086 as shown in SEQ ID NO:
- 10;
- and
- (b) recovering and purifying from said culture medium a protein comprising the amino acid sequence from amino acid #1 to amino acid #109 as shown in SEQ ID NO:

A pharmaceutical composition comprising an effective amount of a BMP-11 protein in dimeric form with another inhibin-β protein, in admixture with a pharmaceutically acceptable vehicle.
A pharmaceutical composition comprising an effective amount of a BMP-11 protein in dimeric form with an inhibin-α protein, in admixture with a pharmaceutically acceptable vehicle.
A pharmaceutical composition comprising an effective amount of a BMP-11 protein in dimeric form with a BMP protein, in admixture with a pharmaceutically acceptable vehicle.
A pharmaceutical composition comprising an effective amount of a BMP-11 protein in dimeric form, wherein said composition further comprises at least one bone morphogenetic protein.
A chimeric DNA molecule comprising a DNA sequence encoding a propeptide from a member of the TGF-β superfamily of proteins linked in frame to a DNA sequence encoding an BMP-11 polypeptide.
