Abstract
The present invention features a novel p21-activated kinase that interacts with steroid hormone receptors, the steroid hormone receptor interacting p21-activated kinase (PAKSI). In general, the invention provides methods of inhibiting hormone related cancers. More particularly, the present invention relates to inhibiting prostate cancer and breast cancer. The present invention further provides methods of activating the therapeutic effects of steroid hormone receptors, particularly the estrogen receptor. Alternatively, the present invention provides methods of diagnosing steroid hormone receptor-related diseases.
Claims24
A substantially pure nucleic acid molecule encoding a steroid hormone-interacting p21-activated kinase (PAKSI) polypeptide.
A substantially pure PAKSI polypeptide.
A method of inhibiting a prostate tumor said method comprising contacting said tumor with a compound that is capable of inhibiting PAKSI.
A method of inhibiting a breast tumor, said method comprising contacting said tumor with a compound that is capable of inhibiting PAKSI.
A method of inhibiting a steroid hormone receptor, said method comprising administering to an individual a compound that is capable of inhibiting PAKSI.
A method of stimulating a steroid hormone receptor, said method comprising administering to an individual a PAKSI agonist.
A method of stimulating the therapeutic effects of estrogen, said method comprising administering to an individual a PAKSI agonist.
A method of inhibiting cardiovascular disease, said method comprising administering to an individual a PAKSI agonist.
A therapeutic composition comprising PAKSI polypeptide formulated in a physiologically acceptable carrier.
A method of determining whether a compound is a PAKSI modulatory compound, said method comprising
- (a) providing a cell expressing a PAKSI polypeptide;
- (b) contacting said cell with said compound;
- and
- (c) measuring the expression or enzymatic-activity of PAKSI by said cell, an alteration in the level of said expression or activity indicating that said compound is a PAKSI modulatory compound.
A method of determining whether a compound is a PAKSI modulatory compound, said method comprising
- (a) providing a PAKSI polypeptide or a PAKSI polypeptide fragment having PAKSI enzymatic activity;
- (b) contacting said PAKSI polypeptide or a PAKSI polypeptide fragment with said compound;
- and
- (c) measuring the enzymatic-activity of PAKSI, an alteration in the level of said activity indicating that said compound is a PAKSI modulatory compound.
A method of diagnosing a mammal for the presence of prostate cancer or increased likelihood of developing prostate cancer, said method comprising measuring PAKSI gene expression in a sample from said mammal, an alteration in said expression relative to a sample from an unaffected mammal being an indication that said mammal has prostate cancer or an increased likelihood of developing prostate cancer.
A method of diagnosing a mammal for the presence of breast cancer or increased likelihood of developing breast cancer, said method comprising measuring PAKSI gene expression in a sample from said mammal, an alteration in said expression relative to a sample from an unaffected mammal being an indication that said mammal has breast cancer or an increased likelihood of developing breast cancer.
A method of diagnosing a mammal for the presence of prostate cancer or increased likelihood of developing prostate cancer, said method comprising measuring PAKSI polypeptide enzymatic activity in a sample from said mammal, an alteration in said activity relative to a sample from an unaffected mammal being an indication that said mammal has prostate cancer or an increased likelihood of developing prostate cancer.
A method of diagnosing a mammal for the presence of breast cancer or increased likelihood of developing breast cancer, said method comprising measuring PAKSI polypeptide enzymatic activity in a sample from said mammal, an alteration in said activity relative to a sample from an unaffected mammal being an indication that said mammal has breast cancer or an increased likelihood of developing breast cancer.
A kit for diagnosing a mammal for the presence of a steroid hormone receptor-related disease.
A method of treating a patient with a disease characterized by abnormal cell growth, said method comprising administering to said patient a PAKSI modulatory compound.
A method of treating a patient with a disease characterized by an abnormal cytoskeleton in a specific cell type, said method comprising administering to said patient a PAKSI modulatory compound.
