Abstract
A protein kinase having a structure related to that of a kinase enzyme of the p70S6K signalling pathway is described, wherein a serine or threonine residue homologous to T389 in p70S6K is replaced with an acidic amino acid residue in order to confer constitutive activity.
Claims11
A protein kinase having a structure related to that of a kinase enzyme of the p70S6K signalling pathway wherein a serine or threonine residue homologous to T389 in p70S6K is replaced with an acidic amino acid residue in order to confer constitutive activity.
A protein kinase according to any preceding claim, wherein threonine residues are replaced with glutamic acid and serine residues are replaced with aspartic acid.
A method for screening compounds for activity as inhibitors of a protein kinase related to a kinase enzyme of the p70S6K signalling pathway comprising assaying for dephosphorylation of the T389 homologous site in said protein kinase in response to the compounds.
A dominant negative mutant of a protein kinase having a structure related to that of a kinase enzyme of the p70S6K signalling pathway wherein T229 is mutated to Alanine.
A method for identifying a kinase enzyme capable of activating a p70S6K-related kinase, comprising assaying a compound or mixture of compounds for ability to activate a mutant of a p70S6K-related kinase.
