Abstract
A microorganism double-stained with appropriate fluorescent reagents is dropped onto a light permeable plate or passed through a micro-tube and irradiated with excitation light. As a result, the fluorescent reagents absorb the excitation light and thus the microorganism fluoresces. This fluorescence is detected by a CCD camera, a photodiode or a multiplier photo-tube and subjected to image-processing, thereby determining the amount and type of the microorganism and judging whether it has vital cells or dead cells.
Claims13
A method for immediately determining microorganisms being characterized in that:
- microorganisms, which are double-stained with appropriate fluorescent reagents, are dropped on a light permeable plate which is processed so as to scatter light;
- an excited light having a central wavelength of 488 nm is illuminated to said microorganisms on the light permeable plate from underneath to make the microorganisms fluoresce;
- and an image of the fluorescent microorganism is picked up by an image pick up means from an upper side;
- wherein an amount of the microorganisms, a type of the microorganisms, and whether the microorganisms are vital or dead are determined.
A method for immediately determining microorganisms being characterized in that:
- microorganisms, which are double-stained with appropriate fluorescent reagents, pass through a micro-tube having a light permeability;
- an excited light having a central wavelength of 488 nm is illuminated to said microorganisms passing through said micro-tube to make the microorganisms emit;
- and the fluoresce of the microorganism is detected by photodiodes and the image of the fluoresce is converted into electric signals;
- wherein an amount of the microorganisms, a type of the microorganisms, and whether the microorganisms are vital or dead are determined.
