Abstract
The present invention relates to compositions and methods relating to the characterization of gene function. Specifically, the present invention provides transgenic mice comprising disruption in a CTSG. The present invention also provides methods of identifying agents that modulate CTSG expression and function, useful models, and potential treatments for various disease states and disease conditions.
Claims31
A transgenic mouse comprising a disruption in a CTSG gene.
A transgenic mouse comprising a disruption in a CTSG gene, wherein there is no native expression of endogenous CTSG gene.
A method of producing a transgenic mouse comprising a disruption in a CTSG gene, the method comprising:
- (a) providing a murine stem cell comprising a disruption in a CTSG gene;
- and
- (b) introducing the murine stem cell into a pseudopregnant mouse, wherein the pseudopregnant mouse gives birth to a transgenic mouse.
A targeting construct comprising:
- (a) a first polynucleotide sequence homologous to at least a first portion of a CTSG gene;
- (b) a second polynucleotide sequence homologous to at least a second portion of a CTSG gene;
- and
- (c) a selectable marker.
A cell comprising a disruption in a CTSG gene.
A method of identifying an agent useful as a treatment for inflammatory bowel disease, the method comprising:
- (a) contacting a test agent with CTSG;
- and
- (b) determining whether the agent modulates CTSG.
A method of identifying an agent useful as a treatment for arthritis, the method comprising:
- (a) contacting a test agent with CTSG;
- and
- (b) determining whether the agent modulates CTSG.
A method of evaluating a potential therapeutic agent capable of affecting a condition associated with a mutation in a CTSG gene, the method comprising:
- (a) administering the potential therapeutic agent to a transgenic mouse comprising a disruption in a CTSG gene;
- and
- (b) evaluating the effects of the agent on the transgenic mouse.
A method of evaluating a potential therapeutic agent capable of affecting a condition associated with a mutation in a CTSG gene, the method comprising:
- (a) contacting the potential therapeutic agent with CTSG;
- (b) evaluating the effects of the agent on the CTSG.
A therapeutic agent for treating inflammatory bowel disease, wherein the agent modulates CTSG.
A therapeutic agent for treating inflammatory bowel disease, wherein the agent is an antagonist of CTSG.
A therapeutic agent for treating arthritis, wherein the agent modulates CTSG.
A therapeutic agent for treating rheumatoid arthritis, wherein the agent is an antagonist of CTSG.
A pharmaceutical composition comprising CTSG.
A method of preparing a pharmaceutical composition for a condition associated with a function of CTSG, the method comprising:
- (a) identifying a compound that modulates CTSG;
- (b) synthesizing the identified compound;
- and
- (c) incorporating the compound into a pharmaceutical carrier.
A method of treating inflammatory bowel disease, the method comprising administering to a subject in need a therapeutically effective amount of an agent that modulates CTSG.
A method of treating rheumatoid arthritis, the method comprising administering to a subject in need a therapeutically effective amount of an agent that modulates CTSG.
Phenotypic data associated with a transgenic mouse comprising a disruption in a CTSG gene, wherein the phenotypic data is in an electronic database.
