Abstract
A conjugate exhibiting interferon gamma activity and comprising at least one first non-polypeptide moiety covalently linked to an IFG polypeptide, the polypeptide comprising an amino acid sequence that differs from that of a parent IFNG polypeptide in at least one introduced and/or at least one removed amino acid residue comprising an attachment group for the non-polypeptide moiety. The conjugate may be used for treatment of various diseases.
Claims35
A conjugate exhibiting IFNG activity and comprising at least one first non-polypeptide moiety covalently attached to an IFNG polypeptide, the polypeptide comprising an amino acid sequence that differs from that of a parent IFNG polypeptide in at least one introduced and/or at least one removed amino acid residue comprising an attachment group for the non-polypeptide moiety.
The conjugate according to claim 17, wherein the second non-polypeptide moiety is a polymer.
A conjugate exhibiting IFNG activity comprising at least one N-terminally PEGylated IFNG polypeptide.
The conjugate according to any of the preceding claims, which has increased functional in vivo half-life as compared to huIFNG.
A method of increasing the functional in vivo half-life of an IFNG polypeptide, which method comprises introducing an amino acid residue constituting an attachment group for a non-polypeptide moiety into a position of a parent IFNG polypeptide comprising a surface exposed amino acid residue which does not contain such attachment group and/or removing an amino acid residue constituting such attachment group, and subjecting the resulting modified polypeptide to conjugation with the non-polypeptide moiety which has the amino acid residue having been introduced and/or removed as an the attachment group.
