Abstract
The discriminating capability of hybridization assays is increased by a combination of labelled primers which produce amplificates of one strand of a nucleic acid with a capture probe which is complementary to the same strand of the nucleic acid.
Claims14
A method for the detection of a nucleic acid comprising the steps:
- a) amplifying the nucleic acid or of a part thereof in a reaction mixture with two primers, one of which can hybridize with one strand of the nucleic acid to be detected and the other of which can hybridize with a strand complementary thereto, to form extension products which are complementary to each other, wherein at least one of the primers contains a detectable label;
- b) contacting a capture probe with the extension products to form a hybridization complex comprising the capture probe and the extension products;
- c) separating the hybridization complex from other components of the reaction mixture;
- and
- d) determining the detectable label in the hybridization complex,
- wherein the capture probe is selected such that it can bind to one of the extension products and said extension product can also hybridize with the extension product formed by extension of the primer containing the detectable label.

A method for the selective detection of specific sequences in one or several nucleic acids(s) from a sample comprising the steps:
- a) amplifying the nucleic acids or parts thereof in a reaction mixture with at least one set of two primers, wherein one primer in each set of primers can hybridize with one strand of the nucleic acids and the other primer in each set can hybridize with a strand complementary thereto, to form extension products which are complementary to each other for each of the specific sequences, wherein at least one primer in each set of primers contains a detectable label;
- b) contacting at least one capture probe with the extension products to form hybridization complexes comprising a capture probe and the extension products,
- c) separating the hybridization complexes from other components of the reaction mixture;
- and
- d) determining the detectable label in the hybridization complexes,
- wherein the capture probes are selected such that they each can bind to one of the extension products and said extension product can also hybridize with the extension product formed by extension of the primer containing the detectable label.
3 more elements
A reagent kit for the detection of nucleic acids containing in separate containers:
- a) at least one primer which contains a detectable label for the amplification of nucleic acids or parts thereof;
- and
- b) at least two immobilizable or immobilized capture probes with different sequences,
- wherein the primer is selected such that it can hybridize with the same strand of an extension product as the capture probes.
