Abstract
The present invention relates to oligonucleotides for use in amplifying and detecting HIV nucleic acid in a sample.
Claims80
An oligonucleotide probe, wherein the base sequence of said probe consists of or is contained within a base sequence perfectly complementary to an HIV-derived target sequence selected from the group consisting of SEQ ID NO:1, SEQ ID NO:4, SEQ ID NO:7, SEQ ID NO:10, SEQ ID NO:13, and the sequences perfectly complementary thereto, wherein said probe hybridizes to HIV-derived nucleic acid in a sample to a form a detectable probe:target duplex which indicates the presence of HIV nucleic acid under hybridization conditions, and wherein said probe does not hybridize to human nucleic acid in said sample to form a detectable probe:non-target duplex under said conditions.
The method of claim 34, wherein said amplifying step is carried out in the presence of reverse transcriptase.
An oligonucleotide, wherein the base sequence of said oligonucleotide consists of or is contained within a base sequence perfectly complementary to an HIV-derived target sequence selected from the group consisting of SEQ ID NO:1, SEQ ID NO:2, SEQ ID NO:3, SEQ ID NO:4, SEQ ID NO:5, SEQ ID NO:6, SEQ ID NO:7, SEQ ID NO:8, SEQ ID NO:9, SEQ ID NO:10, SEQ ID NO:11, SEQ ID NO:12, SEQ ID NO:13, SEQ ID NO:14, SEQ ID NO:15, SEQ ID NO:16, SEQ ID NO:17, SEQ ID NO:18, SEQ ID NO:19, SEQ ID NO:20, SEQ ID NO:21, SEQ ID NO:22, and the sequences perfectly complementary thereto, wherein said oligonucleotide hybridizes to HIV-derived nucleic acid in a sample to form an oligonucleotide:target duplex under hybridization conditions, and wherein said oligonucleotide does not hybridize to human nucleic acid in said sample to form an oligonucleotide:non-target duplex under said conditions.
A method of detecting HIV nucleic acid which may be present in a sample comprising the steps of:
- a) providing to a sample one or more oligonucleotide primers under conditions such that one or more of said primers forms a complex with an HIV-derived target nucleic acid, thereby permitting DNA synthesis to be initiated, wherein the base sequence of each said primer consists of or is contained within a base sequence perfectly complementary to an HIV-derived first target sequence selected from the group consisting of SEQ ID NO:2, SEQ ID NO:3, SEQ ID NO:5, SEQ ID NO:6, SEQ ID NO:8, SEQ ID NO:9, SEQ ID NO:11, SEQ ID NO:12, SEQ ID NO:14, SEQ ID NO:15, SEQ ID NO:16, SEQ ID NO:17, SEQ ID NO:18, SEQ ID NO:19, SEQ ID NO:20, SEQ ID NO:21, SEQ ID NO:22, and the sequences perfectly complementary thereto;
- b) amplifying a second target sequence contained in said HIV-derived target nucleic acid;
- and
- c) detecting amplified product from step b) as an indication of the presence HIV nucleic acid in said sample.
