Abstract
Markers associated with Sclerotinia stem rot resistance are provided. Methods of identifying resistant, and susceptible plants, using the markers are provided. Methods for identifying and isolating QTL are a feature of the invention, as are QTL associated with Sclerotinia stem rot resistance.
Claims22
A method of identifying a Sclerotinia stem rot resistant soybean by identifying a quantitative trait locus (“QTL”) associated with Sclerotinia stem rot resistance, the method comprising:
- (i) detecting at least one nucleic acid from the soybean, which nucleic acid localizes to a chromosome interval flanked on each side by loci having at least about 80% sequence identity to one or more marker pairs selected from among:
- a) Satt155 and SLS1C.L24;
- b) Sat—129 and Satt329;
- c) Satt556 and P1694;
- d) PHP8701R and Satt311;
- e) PHP10118C and Satt231;
- f) P1047 and A724—1;
- and,
- g) Satt523 and SLS2C.F20;
- and,
- (ii) identifying the soybean comprising the nucleic acid, thereby identifying the Sclerotinia stem rot resistant soybean.
A method of positional cloning of a nucleic acid, the method comprising:
- (i) providing a nucleic acid from a soybean, which nucleic acid localizes to a chromosome interval flanked on each side by loci having at least about 80% sequence identity to one or more marker pairs selected from among:
- a) Satt155 and SLS1C.L24;
- b) Sat—129 and Satt329;
- c) Satt556 and P1694;
- d) PHP8701R and Satt311;
- e) PHP10118C and Satt231;
- f) P1047and A724—1;
- and,
- g) Satt523 and SLS2C.F20;
- and,
- (ii) cloning the nucleic acid.
An isolated or recombinant nucleic acid comprising:
- a) a polynucleotide sequence selected from:
- SEQ ID NO:
- 1 (SLS1C.L24), SEQ ID NO:
- 2 (SLS2C.F20) and SEQ ID NO:
- 3 (PHP8701R);
- b) a polynucleotide sequence with at least about 80% sequence identity to a polynucleotide sequence of (a), and/or
- c) a polynucleotide sequence complementary to (a) or (b).
