Abstract
Human EMR2 (EGF-like molecule containing mucin-like hormone receptor-2) a polypeptide of SEG ID NO. 2, and fragments and variants thereof, is a member of the EGF-TM7 family and is useful in the control of wound healing and other conditions associated with neutrophilis and macrophages.
Claims41
The use of an EMR2 polypeptide-type polypeptide in the preparation of a medicament for use in a method of therapy of a condition or disease associated with an interaction between the EMR2 polypeptide and an EMR2 ligand, wherein the EMR2 polypeptide-type polypeptide comprises a domain corresponding to the EGF4 domain of EMR2, and which binds to an antibody specific to the EGF4 domain.
The use of an EMR2 polypeptide-type polypeptide in the preparation of a diagnostic agent for use in a method of diagnosis of a condition disease associated with an interaction between the EMR2 polypeptide and an EMR2 ligand, wherein the EMR2 polypeptide-type polypeptide comprises a domain corresponding to the EGF4 domain of EMR2, and which binds to an antibody specific to the EGF4 domain.
A use according to any preceding claim, wherein the EMR2 polypeptide-type polypeptide comprises all or part of the sequence of SEQ ID NO. 2, or a variant thereof.
A use according to any preceding claim, wherein the EMR2 polypeptide-type polypeptide comprises all or part of the sequence of SEQ ID NO. 20, or a variant thereof.
A use according to any preceding claim, wherein the EMR2 polypeptide-type polypeptide comprises up to 15 EGF subunits.
A use according to any preceding claim, wherein the EMR2 polypeptide-type polypeptide comprises up to 10 EGF subunits.
A use according to any preceding claim, wherein the EMR2 polypeptide-type polypeptide comprises 5 EGF subunits.
A use according to any preceding claim, wherein the EMR2 polypeptide-type polypeptide comprises at least one EGF subunit, and wherein at least one EGF subunit corresponds to EGF subunit 4 of naturally occurring EMR2, or a variant thereof.
A use according to any preceding claim, wherein the EMR2 polypeptide-type polypeptide substantially lacks any transmembrane sequences.
A use according to any preceding claim, wherein the polypeptide-type polypeptide is soluble in saline.
A use according to any preceding claim, wherein the EMR2 polypeptide-type polypeptide is substantially identical with the sequence of SEQ ID NO. 2, where the sequences correspond.
A use according to any preceding claim, wherein the EMR2 polypeptide-type polypeptide is not glycosylated.
An agonist for the polypeptide of SEQ ID NO. 2 or 20.
An antagonist for the polypeptide of SEQ ID NO. 2 or 20.
An antibody specific for the polypeptide of SEQ ID NO. 2 or 20.
An antibody specific for the polypeptide of SEQ ID NO. 2 or 20 which is a monoclonal antibody.
A polynucleotide, or a variant thereof, encoding an EMR2 polypeptide-type polypeptide which is
- EMR2 polypeptide protein comprising the amino acid sequence of SEQ ID NO. 2 or SEQ ID NO. 20;
- a polypeptide which has substantial identity therewith and which retains an EMR2 polypeptide biological activity;
- or
- a fragment of such an EMR2 polypeptide protein or substantially identical polypeptide, and which fragment retains an EMR2 polypeptide biological activity.
A method for the diagnosis of EMR2 expression comprising detecting EMR2 in a sample with an antibody specific for EMR2 and assaying the extent of binding of said antibody.
A pharmaceutical composition comprising
- an EMR2 polypeptide-type polypeptide which is
- EMR2 polypeptide protein comprising the amino acid sequence of SEQ ID NO. 2 or 20;
- a polypeptide which has substantial identity therewith and which retains an EMR2 polypeptide biological activity;
- or
- a fragment of such an EMR2 polypeptide protein or substantially identical polypeptide, and which fragment retains an EMR2 polypeptide biological activity;
- in the preparation of a diagnostic agent for use in a method of diagnosis of a condition or disease associated with EMR2 polypeptide.
