An assay method comprising the detection of holotranscobalamin II (holo-TCII) and folate in a cobalamin-containing sample from a subject.

An assay method comprising the detection of holo-transcobalamin II (holo-TCII) and folate in a cobalamin-containing sample from a subject wherein said detection of holo-TCII comprises the step of contacting said sample with a specific binding ligand for an analyte chosen from the set of TCII and holo-TCII and separating the ligand bound fraction from the non ligand bound fraction whereby to concentrate said analyte by at least 3 fold to give a concentrated sample, followed by determination of the holo-TCII level.
