Abstract
Combinatorial libraries comprise first oligonucleotide analogs and second oligonucleotide analogs which are coupled together to form antisense molecules capable of binding target polynucleotides and activating an RNase, and ribozymes capable of cleaving polynucleotides.
Claims39
A method of increasing the effectiveness of an antisense oligonucleotide to cleave a target molecule, comprising:
- providing an antisense oligonucleotide comprising an RNase H activation region;
- and
- adding at least two adjacent universal bases to said oligonucleotide, wherein the addition of said universal bases increases the activity of RNAase H to cleave a target molecule.
A method of cleaving a target RNA molecule, comprising:
- providing an antisense oligonucleotide comprising an RNAase H activation region;
- adding at least two adjacent universal bases to said oligonucleotide, wherein the addition of said universal bases increases the activity of RNAase H to cleave a target RNA molecule;
- and
- contacting said target RNA molecule with said antisense oligonucleotide in the presence of RNase H.

A method of making an antisense oligonucleotide that cleaves a target molecule, comprising:
- providing an antisense oligonucleotide comprising an RNAase H activation region;
- and
- adding at least two adjacent universal bases to said oligonucleotide, wherein the addition of said universal bases increases the activity of RNAase H to cleave a target molecule.
