Abstract
The present invention relates to DNA molecules and DNA constructs which promote papaya fruit ripening. The present invention is also directed to methods for promoting or delaying the ripening of papaya plants through transformation of papaya with DNA constructs containing nucleic acids which encode proteins or polypeptides involved in papaya ripening. The invention also relates to expression systems, host cells, and plants containing such DNA constructs.

An isolated nucleic acid molecule encoding a protein or polypeptide which controls papaya fruit ripening, wherein the protein or polypeptide is a β-galactosidase, and the nucleic acid molecule either:
- (a) has a nucleotide sequence of SEQ ID NO:
- 1, (b) encodes a protein having an amino acid sequence of SEQ ID NO:
- 2, or (c) hybridizes to at least 20 nucleotides of the DNA molecule having a nucleotide sequence of SEQ ID NO:
- 1 under stringent conditions characterized by a hybridization buffer comprising 5×SSC buffer at a temperature of 45° C.
An isolated nucleic acid molecule encoding a protein or polypeptide which controls papaya fruit ripening, wherein the protein or polypeptide is a β-galactosidase, and the nucleic acid molecule either:
- (a) has a nucleotide sequence of SEQ ID NO:
- 3, (b) encodes a protein having an amino acid sequence of SEQ ID NO:
- 4, or (c) hybridizes to at least 20 nucleotides of the DNA molecule having a nucleotide sequence of SEQ ID NO:
- 3 under stringent conditions characterized by a hybridization buffer comprising 5×SSC buffer at a temperature of 45° C.
A DNA construct comprising:
- one or more operatively linked nucleic acid molecules, wherein the nucleic acid molecule encodes a protein or polypeptide which controls papaya fruit ripening;
- an operably linked heterologous DNA promoter, and
- an operably linked 3′ regulatory region.
