Abstract
The invention features a method for triggering release of a drug from a hydrogel polymer to tissue at a desired location of the body using a catheter. A portion of the catheter is coated on its outer surface with a polymer having the capacity to incorporate a predetermined substantial amount of drug which is immobilized in the polymer until released by a triggering agent or condition that is different from physiological conditions. Upon contact with a triggering agent or condition, the polymer reacts, e.g., swells or contracts, such that the drug is delivered to the desired body tissue. A balloon catheter is shown in which the hydrogel is carried on the exterior surface of the balloon.
Claims23
A method for delivering a drug to tissue at a desired location within the body, comprising:
- providing a catheter constructed for insertion in the body, a portion of said catheter carrying a hydrogel having the capacity to incorporate a predetermined substantial amount of said drug, said drug being substantially immobilized in said hydrogel until released by a triggering agent or condition that differs from ambient physiological conditions;
- introducing said catheter to said body to the point of desired drug application;
- and
- exposing said hydrogel to said triggering agent or condition, wherein said triggering agent or condition triggers release of said drug from said hydrogel for delivery to said desired location within the body.
A kit comprising (a) a catheter constructed for insertion in a body lumen having a catheter shaft having an expandable balloon portion mounted on said catheter shaft, said balloon portion carrying a hydrogel polymer having the capacity to incorporate a predetermined substantial amount of a drug;
- (b) said drug, wherein said drug is capable of being immobilized in said polymer until released by a triggering agent;
- and (c) a solution of said triggering agent.
A kit comprising (a) a thermal catheter constructed for insertion in a body lumen having a catheter shaft having an expandable balloon portion mounted on said catheter shaft, said balloon portion carrying a hydrogel polymer having the capacity to incorporate a predetermined substantial amount of a drug;
- (b) said drug, wherein said drug is capable of being immobilized in said polymer until released by an increase in temperature of said polymer.
A kit comprising (a) a catheter constructed for insertion in a body lumen having a catheter shaft having an expandable balloon portion mounted on said catheter shaft and a fiber capable of emitting electromagnetic radiation to said balloon portion, said balloon portion carrying a polymer having the capacity to incorporate a predetermined substantial amount of a drug;
- (b) said drug, wherein said drug is capable of being immobilized in said polymer until released by exposure to electromagnetic radiation.
A catheter for delivering a drug to tissue at a desired location in the body, comprising:
- a catheter constructed for insertion in a body lumen having a catheter shaft with an expandable balloon portion mounted on said catheter shaft;
- said balloon portion having pores through which a triggering agent can pass to its outer surface;
- said balloon portion being expandable to engage said tissue at a controlled pressure to fill a cross-section of the body lumen and press against said wall of said body lumen;
- said balloon portion being coated on its outer surface with a hydrogel polymer having the capacity to incorporate a predetermined substantial amount of said drug, said drug being immobilized in said polymer until released by a triggering agent, whereby release of said drug from said polymer is triggered by contacting said polymer with said triggering agent delivered through said pores.
A catheter for delivering a drug to tissue at a desired location of the wall of a body lumen, comprising:
- a catheter constructed for insertion in a body lumen having a catheter shaft having an expandable balloon portion mounted on said catheter shaft;
- said balloon portion being expandable to engage said tissue at a controlled pressure to fill the cross-section of the body lumen and press against the wall of said body lumen;
- a porous membrane positioned over at least a portion of the surface of said balloon portion and creating therebetween a space into which a triggering agent is introduced;
- said porous membrane being coated on its outer surface with a hydrogel polymer having the capacity to incorporate a predetermined substantial amount of said drug, said drug being immobilized in said polymer until released by a triggering agent and said porous membrane being constructed to transmit said triggering agent from said space through openings in said membrane to the outer surface of said membrane in response to pressure applied by inflation of said balloon portion to compress said space, whereby expanding said balloon portion forces said triggering agent into said polymer and triggers release of said drug at said desired location of the wall of said body lumen.
A catheter for delivering a drug to tissue at a desired location in the body, comprising:
- a thermal catheter constructed for insertion in a body lumen having a catheter shaft with an expandable balloon portion mounted on said catheter shaft;
- said balloon portion being expandable to engage said tissue at a controlled pressure to fill a cross-section of the body lumen and press against said wall of said body lumen;
- said balloon portion being coated on its outer surface with a temperature-sensitive hydrogel polymer having the capacity to incorporate a predetermined substantial amount of said drug, said drug being immobilized in said polymer until released by an increase in temperature of said polymer, whereby release of said drug from said polymer is triggered by an increase in temperature of said polymer.
A catheter for delivering a drug to tissue at a desired location in the body, comprising:
- a catheter constructed for insertion in a body lumen having a catheter shaft with an expandable balloon portion mounted on said catheter shaft;
- said catheter having a fiber capable of emitting electromagnetic radiation to said balloon portion;
- said balloon portion being expandable to engage said tissue at a controlled pressure to fill a cross-section of the body lumen and press against said wall of said body lumen;
- said balloon portion being coated on its outer surface with an electromagnetic radiation-sensitive hydrogel polymer having the capacity to incorporate a predetermined substantial amount of said drug, said drug being immobilized in said polymer until released by exposure to said electromagnetic radiation, whereby release of said drug from said polymer is triggered by exposing said polymer to said electromagnetic radiation.
