Abstract
Compositions and methods are provided for the enhanced in vitro synthesis of nucleic acids. Nucleoside monophosphates are converted to nucleoside triphosphates by in situ reactions to continuously supply the required nucleoside triphosphates. This approach provides for more homeostatic reaction conditions and lower costs.
Claims11
A method for in vitro synthesis of nucleic acids in a reaction mix, the method comprising:
- generating nucleoside triphosphates from nucleoside monophosphates by catalysis with nucleoside monophosphate kinase, nucleoside diphosphate kinase and ATP;
- generating ATP from an energy source using an ATP generating enzyme;
- and
- synthesizing said nucleic acids in said reaction mix utilizing said nucleoside triphosphates.
