Abstract
An isolated polypeptide comprising an amino acid sequence having at least 80% sequence identity to the sequence SEQ ID NOS:4 or 6, polynucleotides encoding these peptides, and antibodies to the polypeptides are useful in treating metabolic disorders or disorders associated with changes in adipose tissue physiological function or mass.
Claims53
An isolated polypeptide comprising an amino acid sequence having at least 80% sequence identity to the sequence of SEQ ID NOS:4 or 6.
An isolated polynucleotide comprising a nucleotide sequence having at least 80% sequence identity to the sequence of SEQ ID NOS:1, 2, 3 or 5, or a complement of said polynucleotide.
A method of treating metabolic disorders comprising modulating the activity of AAP.
A method of detecting a metabolic disorder, or a disorder associated with changes in adipose tissue physiological function or mass, comprising:
- detecting a change in expression or activity of AAP.
A method for determining whether a compound up-regulates or down-regulates the transcription of an AAP gene, comprising:
- contacting said compound with a composition comprising a RNA polymerase and said gene and measuring the amount of AAP gene transcription.
A method for determining whether a compound up-regulates or down-regulates the translation of an AAP gene, comprising:
- contacting said compound with a composition comprising a ribosome and a polynucleotide corresponding to a mRNA of said gene and measuring the amount of AAP gene translation.
A method of screening a tissue sample for the presence of cells likely to differentiate into adipocytes, comprising:
- measuring expression of AAP in said tissue sample.
A transgenic non-human animal, having a disrupted AAP gene.
A transgenic non-human animal, comprising an exogenous polynucleotide having at least 80% sequence identity to the sequence of SEQ ID NOS:1, 2, 3 or 5, or a complement of said polynucleotide.
A method of screening a sample for an AAP gene mutation, comprising:
- comparing an AAP nucleotide sequence in the sample with SEQ ID NOS:4 or 6.
A method of determining if a cell is likely to become an adipocyte, comprising:
- measuring expression of AAP in said cell.
A method of inhibiting adipocyte differentiation, comprising inhibiting the activity of AAP.
A method to measuring AAP agonist or antagonist activity of a compound comprising:
- contacting the compound with a composition comprising a polypeptide having at least 80% sequence identity to the sequence of SEQ ID NO:
- 4 or SEQ ID NO:6 and determining if AAP activity is changed.
