Abstract
A method for influencing the separation mobility of single stranded nucleic acids in native conditions is disclosed. Temperature changes during the separation of single stranded nucleic acids under native conditions are used as the basis of a novel method of detection of specific nucleic acid sequences.
Claims56
A method for detecting a variation in a nucleic acid molecule said method comprising:
- (a) providing said nucleic acid;
- (b) transforming said nucleic acid into one or more single stranded nucleic acid spatial conformer(s);
- (c) separating said conformer(s) under native conditions;
- (d) changing one or more conditions of said separation at least one time during said separation wherein said change can impart a conformation change in said conformer;
- (e) detecting the mobility pattern of said conformer(s);
- and
3 more elements
A method for detecting variation in a population of nucleic acid molecules said method comprising:
- (a) providing said population of nucleic acid molecules;
- (b) transforming said nucleic acid molecules into one or more single stranded nucleic acid spatial conformer(s);
- (c) separating said conformer(s) under native conditions;
- (d) changing one or more conditions of said separation at least one time during said separation wherein said change can impart a conformation change in said conformer.
- (e) detecting the mobility pattern of said conformer(s).

A system comprising:
- (a) a holding structure that holds a separation gel;
- (b) a heat exchange module through which a cooling medium flow, said heat exchange module being coupled to said holding structure;
- (c) input devices coupled to opposite ends of said gel and to a source;
- (d) a sensor that detects a temperature of said gel or said cooling medium;
- and
- (e) a control system that receives signals from said sensor so that said gel is maintained at two or more temperatures during two or more corresponding time intervals during a separation under native conditions.
