Abstract
The present invention relates to the field of immunology and hyperproliferative diseases. More specifically, the present invention relates to a method of detecting and monitoring therapeutic antibody:antigen complex, soluble antigen and soluble therapeutic antibody, wherein a patient has undergone at least one course of immunotherapy. Yet further, levels of therapeutic antibody:antigen complexes, soluble antigens or soluble therapeutic antibodies may be measured and used to stage or monitor a hyperproliferative disease.
Claims73
A method of determining the efficacy of an antibody immunotherapy in a patient comprising detecting the levels of circulating antibody:antigen complexes.
A method of an immunotherapy comprising administering to a patient a therapeutic antibody and detecting the presence of a circulating antibody:antigen complex.
A method of detecting a therapeutic antibody:antigen complex comprising:
- obtaining a sample from a patient, wherein the patient has undergone a course of immunotherapy with a therapeutic antibody in which the therapeutic antibody binds to a soluble circulating target antigen forming the complex;
- contacting the sample with a first monoclonal antibody, wherein the antibody captures the complex;
- contacting the complex with a labeled second antibody;
- and
- measuring the labeled complex.
A method of monitoring a therapeutic antibody therapy comprising measuring a therapeutic antibody:antigen complex in a patient sample.
A method of monitoring a hyperproliferative disease comprising measuring a therapeutic antibody:antigen complex in a patient sample.
A method of monitoring an autoimmune disease comprising measuring a therapeutic antibody:
- antigen complex in the patient serum.
A method of determining tumor mass comprising:
- obtaining a sample from a patient;
- contacting the sample with a first monoclonal antibody, wherein the antibody captures a soluble antigen in the sample;
- contacting the soluble antigen with a labeled second antibody;
- and
- measuring the labeled soluble antigen.
A method of staging a B cell hyperproliferative disease comprising determining the level of soluble CD20 in a patient sample by a sandwich ELISA.
A method of monitoring B cell hyperproliferative disease comprising measuring the level of soluble CD20, soluble anti-CD20 or CD20/anti-CD20 complexes in a patient sample by a sandwich ELISA.
A method of monitoring hematopoietic hyperproliferative disease comprising measuring the level of soluble CD52, soluble anti-CD52 and CD52/anti-CD52 complexes by a sandwich ELISA.
A method of staging T cell hyperproliferative disease comprising measuring the level of soluble CD52 in a patient sample by a sandwich ELISA.
A method of monitoring T cell hyperproliferative disease comprising measuring the level of soluble CD52, soluble anti-CD52 or CD52/anti-CD52 complexes in a patient sample by a sandwich ELISA.
A method of staging a myeloid hyperproliferative disease comprising determining the level of soluble CD33 in a patient sample by a sandwich ELISA.
A method of monitoring myeloid hyperproliferative disease comprising measuring the level of soluble CD33, soluble anti-CD33 or CD33/anti-CD33 complexes in a patient sample by a sandwich ELISA.
