Abstract
Ultra high affinity antibodies with binding affinities in the range of 1010 M−1, and even 1011 M−1 are disclosed. Such antibodies include antibodies having novel high affinity complementarity determining regions (CDRs), especially those with framework and constant regions derived from either humans or mice. Methods of preparing and screening such antibodies, as well as methods of using them to prevent and/or treat disease, especially virus-induced diseases, are also disclosed.
Claims46
A high affinity neutralizing immunoglobulin comprising at least three high affinity complementarity determining regions (CDRS) wherein each said high affinity CDR has an amino acid sequence selected to result in an immunoglobulin with specificity toward at least one antigenic determinant and having an affinity constant (Ka) of at least 1010M−1 for said antigenic determinant.
A recombinant high affinity neutralizing immunoglobulin having an affinity constant of at least 1010 M−1, wherein said immunoglobulin comprises a human constant region and a heavy and light chain framework region at least part of which is derived from human antibodies.
