Specialist signal profilers for base calling

A system, comprising:

• a sequencing instrument;
• at least one processor;
• and
• a non-transitory computer-readable medium comprising instructions thereon that, when executed by the at least one processor, cause the system to:
• capture, by one or more light detectors of the sequencing instrument at imaging events of sequencing cycles of a sequencing run, per-cycle sequencing image sets comprising signals from analytes:
• fit a plurality of signal distributions on initially sequenced signals detected during initial sequencing cycles of the sequencing run;
• separately train respective specialist signal profilers in a plurality of specialist signal profilers (i) to maximize signal-to-noise ratio of respective signal distributions in the plurality of signal distributions (ii) using intensity data of the initially sequenced signals segmented according to the plurality of signal distributions;
• store the trained respective specialist signal profilers in memory;
• and
• execute a base calling operation by:
• uniquely mapping subsequently sequenced signals detected during subsequent sequencing cycles of the sequence run to the respective signal distributions;
• applying the trained respective specialist signal profilers stored in memory to the subsequently sequenced signals segmented based on the unique mapping to the respective signal distributions;
• and
• generating base calls for the subsequent sequencing cycles using respective noise-corrected intensity data output by the trained respective specialist signal profilers.

A system, comprising:

• a sequencing instrument;
• at least one processor;
• and
• a non-transitory computer-readable medium comprising instructions thereon that, when executed by the at least one processor, cause the system to:
• capture, by one or more light detectors of the sequencing instrument at imaging events of sequencing cycles of a sequencing run, per-cycle sequencing image sets comprising signals from a population of analytes;
• process initially sequenced signals detected during initial sequencing cycles of the sequencing run on an analyte-by-analyte basis, to fit respective signal profiles for respective analytes in the population of analytes;
• separately train respective specialist signal profilers in a plurality of specialist signal profilers (i) to maximize signal-to-noise ratio of the respective signal profiles fitted for the respective analytes (ii) using intensity data of the initially sequenced signals segmented according to the respective signal profiles;
• store the trained respective specialist signal profilers in memory;
• uniquely map subsequently sequenced signals detected during subsequent sequencing cycles of the sequence run to the respective signal profiles on the analyte-by-analyte basis;
• apply the trained respective specialist signal profilers stored in memory to the subsequently sequenced signals segmented based on the unique mapping to the respective signal profiles;
• and
• generate base calls for the subsequent sequencing cycles on the analyte-by-analyte basis using respective noise-corrected intensity data output by the trained respective specialist signal profilers.
