Abstract
Described herein is an RNA detection system and methods of using for detecting one or more RNA target molecules. The system comprises an upstream molecular circuitry system comprising one or more RNA toehold switch-based sensors that are activated in the presence of an RNA target molecule to produce a restriction enzyme which cleaves and releases a redox active reporter molecule that binds a capture molecule bound to an electrode, thereby producing a detectable electrochemical signal.
Claims7
An RNA detection system comprising:
- i) an RNA toehold switch-based sensor specific to an RNA target molecule, wherein the RNA toehold switch-based sensor comprises an mRNA sequence encoding a restriction enzyme, such that the restriction enzyme is translated upon binding of the RNA toehold switch-based sensor to the RNA target molecule;
- ii) a reporterDNA/iDNA complex, wherein the reporterDNA is coupled to a redox active molecule, and wherein the restriction enzyme binds and cleaves to said reporterDNA/iDNA complex thereby releasing the reporterDNA;
- and
- iii) a captureDNA and an electrode, wherein the captureDNA is coupled to the electrode and wherein upon release of the reporterDNA, the reporterDNA binds to the captureDNA thereby producing an electrochemical signal detected by the electrode.
