Claims28
An engineered polypeptide comprising:
- an ADAR2 catalytic domain that comprises a sequence that is at least 95% identical to SEQ ID NO:
- 2 and comprises a E488X1 mutation and a N496X2 mutation, wherein the mutations are relative to SEQ ID NO:2, wherein X1 is Q, H, R, K, N, A, M, S, F, L, or W and X2 is F or Y.
An engineered polypeptide comprising:
- an ADAR2 catalytic domain that comprises a sequence that is at least 95% identical to SEQ ID NO:
- 2 from amino acid 316 to amino acid 697 of SEQ ID NO:
- 2 and having a E488X1 mutation and a N496X2 mutation, wherein the mutations are relative to SEQ ID NO:2, wherein X1 is Q, H, R, K, N, A, M, S, F, L, or W and X2 is F or Y.
A composition comprising:
- an engineered polypeptide or a polynucleotide encoding for the engineered polypeptide, wherein the engineered polypeptide comprises:
- an ADAR2 catalytic domain that comprises a sequence that is at least 95% identical to SEQ ID NO:
- 2 from amino acid 316 to amino acid 465 of SEQ ID NO:
- 2;
- a second domain of ADAR2 that comprises a sequence that is at least 95% identical to SEQ ID NO:
- 2 from amino acid 466 to amino acid 701;
- an E488X1 mutation, wherein X1 is Q, H, R, K, N, A, M, S, F, L, or W;
- and
- a N496X2 mutation, wherein X2 is F or Y,
- wherein the mutations are relative to SEQ ID NO:2.
