Abstract
The present disclosure relates to construction of three-dimensional organs from pluripotent stem cells. The present disclosure also relates to a method of forming a cell condensate for self-organization.
Claims18
A method of preparing a cell condensate in vitro, comprising culturing a mixture of cells and/or tissues of a desired type and mesenchymal cells to form a cell condensate,
- wherein the culture is performed on a U- or V-shaped bottom support,
- wherein the support comprises a gel-like support on which the mesenchymal cell is capable of contraction,
- wherein a size of the cell condensate is 1 mm or more, the cells mixed with the mesenchymal cells are pluripotent cells, and/or the tissues mixed with the mesenchymal cells are tissues induced from pluripotent cells, and
- wherein stiffness of a central part of the gel-like support is greater than stiffness of a peripheral part thereof,
- stiffness of a peripheral part of the gel-like support is greater than stiffness of a central part thereof,
- the gel-like support is patterned and has one or more patterns in which stiffness of a central part is greater than stiffness of a peripheral part, or
- the gel-like support is patterned and has one or more patterns in which stiffness of a peripheral part is greater than stiffness of a central part.
A method of preparing a cell condensate in vitro, comprising culturing a mixture of cells and/or tissues of a desired type and mesenchymal cells to form a cell condensate, in which the mesenchymal cells exclude fibroblast,
- wherein the culture is performed on a U- or V-shaped bottom support,
- wherein the support comprises a gel-like support on which the mesenchymal cell is capable of contraction,
- wherein a total cell count of the cells and/or tissues of a desired type is 400,000 or more and a total cell count of the mesenchymal cells are 100,000 to 400,000 in number, or the cells and/or tissues mixed with the mesenchymal cells are derived from liver, pancreas, intestine, lung, kidney, heart, brain or cancer, and
- wherein stiffness of a central part of the gel-like support is greater than stiffness of a peripheral part thereof, stiffness of a peripheral part of the gel-like support is greater than stiffness of a central part thereof, the gel-like support is patterned and has one or more patterns in which stiffness of a central part is greater than stiffness of a peripheral part, or the gel-like support is patterned and has one or more patterns in which stiffness of a peripheral part is greater than stiffness of a central part.
