Abstract
Provided herein are plasma kallikrein antibodies binding to active plasma kallikrein and methods of using such antibodies in preventing hereditary angioedema attack or reducing the rate of hereditary angioedema attack.
Claims17
A method of preventing hereditary angioedema (HAE) attack or reducing the rate of HAE attack, the method comprising:
- administering to a subject in need thereof 300 mg of an antibody that binds to active plasma kallikrein,
- wherein the antibody comprises a heavy chain complementarity determining region (HC CDR) 1 set forth in SEQ ID NO:
- 5, a HC CDR2 set forth in SEQ ID NO:
- 6, a HC CDR3 set forth in SEQ ID NO:
- 7, a light chain CDR (LC CDR) 1 set forth in SEQ ID NO:
- 8, a LC CDR2 set forth in SEQ ID NO:
- 9, and LC CDR3 set forth in SEQ ID NO:
- 10,
- wherein the antibody is formulated in 30 mM sodium phosphate, citric acid, 50 mM histidine, 90 mM sodium chloride, and 0.01% Tween 80, pH 6.0,
- wherein the antibody is administered every two to four weeks for at least two times,
- wherein the subject is a human patient who has experienced at least nine HAE attacks within three months prior to the first administration of the antibody, and
- wherein administration of the antibody reduces cleaved kininogen of the subject to a level comparable to or below the level of healthy subjects.

- 10,
- wherein the antibody is formulated in 30 mM sodium phosphate, citric acid, 50 mM histidine, 90 mM sodium chloride, and 0.01% Tween 80, pH 6.0, and
- the antibody being first administered for a loading period, and then for a maintenance period;
- wherein in the loading period, the antibody is administered once every week, and wherein in the maintenance period, the antibody is administered once every two to four weeks;
- wherein the subject is a human patient who has experienced at least nine HAE attacks within three months prior to the first administration of the antibody;
- and
- wherein administration of the antibody reduces cleaved kininogen of the subject to a level comparable to or below the level of healthy subjects.
