Abstract
The invention provides antibodies that specifically bind sortilin. The antibodies inhibit or delay pathologies associated with changes in progranulin levels and associated symptomatic deterioration.
Claims32
A method of increasing progranulin levels in a subject having or having an increased risk of developing frontotemporal dementia, Alzheimer's disease, Parkinson's disease, amyotrophic lateral sclerosis, or Batten disease, comprising administering to the subject an effective dose of a humanized antibody that specifically binds to human sortilin comprising:
- a humanized heavy chain variable region comprising CDRs of SEQ ID NO:
- 53, SEQ ID NO:
- 54, and SEQ ID NO:
- 55;
- a humanized light chain variable region comprising CDRs of SEQ ID NO:
- 59, SEQ ID NO:
- 72, and SEQ ID NO:
- 61;
- a humanized light chain constant region fused to the humanized light chain variable region;
- and
- a humanized heavy chain constant region, with or without a C-terminal lysine, fused to the humanized heavy chain variable region, wherein the humanized heavy chain constant region comprises:
- (i) an A at position 234, an A at position 235, a Y at position 252, a T at position 254, and an E at position 256, according to EU nomenclature;
- or
- (ii) a Y at position 252, a T at position 254, or an E at position 256, according to EU nomenclature.
A method of treating or effecting prophylaxis of frontotemporal dementia, Alzheimer's disease, Parkinson's disease, amyotrophic lateral sclerosis, or Batten disease in a subject, comprising administering an effective dose of a humanized antibody that specifically binds to human sortilin comprising:
- a humanized heavy chain variable region comprising CDRs of SEQ ID NO:
- 53, SEQ ID NO:
- 54, and SEQ ID NO:
- 55;
- a humanized light chain variable region comprising CDRs of SEQ ID NO:
- 59, SEQ ID NO:
- 72, and SEQ ID NO:
- 61;
- a humanized light chain constant region fused to the humanized light chain variable region;
- and
- a humanized heavy chain constant region, with or without a C-terminal lysine, fused to the humanized heavy chain variable region, wherein the humanized heavy chain constant region comprises:
- (i) an A at position 234, an A at position 235, a Y at position 252, a T at position 254, and an E at position 256, according to EU nomenclature;
- or
- (ii) a Y at position 252, a T at position 254, or an E at position 256, according to EU nomenclature.
