## Abstract

Disclosed herein are peptides having activity as cell penetrating peptides. In some embodiments, the peptides can comprise a cell penetrating peptide moiety and beta-haripin turn creating moiety. In other embodiments, the peptides also comprise a cargo moiety.

## Claims

### Claim 1

A compound comprising;
i. a cyclic cell penetrating 8 amino acid peptide, wherein the amino acids are connected through peptide bonds, that contains;
a. one arginine within two amino acids of another arginine;
b. a phenylalanine within two amino acids of another phenylalanine; and
ii. an exocyclic cargo moiety conjugated to a side chain of the cell penetrating peptide wherein the conjugation is through glutamine and wherein the cargo comprises an amino acid sequence that targets an enzyme.

### Claim 2 (depends on 1)

The compound of claim 1, wherein the cargo moiety also comprises a therapeutic moiety and a linker moiety.

### Claim 3 (depends on 1)

The compound of claim 1, wherein the cargo moiety also comprises a therapeutic moiety and a linker moiety.

### Claim 4 (depends on 1)

The compound of claim 1, wherein the cargo moiety also comprises one or more detectable moieties or one or more therapeutic moieties or any combination thereof.

### Claim 5 (depends on 1)

The compound of claim 1, wherein the cyclic cell penetrating peptide comprises at least three amino acids with a hydrophobic side chain.

### Claim 6 (depends on 5)

The compound of claim 5, wherein at least one of the amino acids with a hydrophobic side chain is a glycine.

### Claim 7 (depends on 1)

The compound of claim 1, wherein the cyclic cell penetrating peptide comprises at least three amino acids which have a hydrophobic side chain, wherein at least two amino acids having a hydrophobic side chain are consecutive.

### Claim 8 (depends on 7)

The compound of claim 7, wherein at least one of the amino acids with a hydrophobic side chain is a glycine.

### Claim 9 (depends on 1)

The compound of claim 1, wherein the cyclic cell penetrating peptide comprises at least two consecutive L amino acids.

### Claim 10 (depends on 1)

The compound of claim 1, wherein each amino acid of the cyclic cell penetrating peptide is a natural amino acid.
