Abstract
The present application provides tricyclic urea compounds that modulate the activity of the V617F variant of JAK2, which are useful in the treatment of various diseases, including cancer.

A compound of Formula Ia:
- or a pharmaceutically acceptable salt thereof, wherein:
- R1 is selected from phenyl and indazolyl, each of which is optionally substituted by 1 or 2 substituents independently selected from C1-6 alkyl and C1-6 alkoxy;
- R2 is C1-6 alkyl;
- R3 is selected from halo and C1-6 alkoxy;
- R4 is C1-6 alkyl;
- and
- R5 is selected from H, halo, C1-3 haloalkyl, and CN;
- wherein each hydrogen atom is optionally replaced by deuterium.
