Abstract
Collection of target analytes from complex samples is performed in detection microwell which includes a size exclusion filter and allows incubating the target analyte with affinity agents for a target analyte for capture and removal of non-target molecules. Detection of the target analytes by collection of the complexes in close proximity to a working electrode and a reference electrode facilitates the detection electrochemical labels produced.
Claims18
A filtering device for filtering target analytes from non-target components comprising:
- an upper reagent well;
- a first layer including first and second surfaces on opposite sides thereof,
- wherein the first and second surfaces define outer edges of the first layer, and
- wherein the first layer has a plurality of holes or openings extending between the first and second surfaces,
- wherein an end of each of the holes or openings of the first layer are in fluid communication with the upper reagent well;
- and
- a second layer coupled to the second surface of the first layer, the second layer including a plurality of size exclusion filters in alignment with the plurality of holes or openings of the first layer, said size exclusion filters including a plurality of pores in alignment with the plurality of holes or openings of the first layer;
- and
- a capillary in fluid communication with a side of the size exclusion filters opposite the upper reagent well.
