Abstract
The present invention provides compositions comprising phase separated nanoparticles, as well as methods of making the nanoparticles and uses thereof. The nanoparticles can be conjugated to therapeutics and used to treat diseases or to screen compounds.
Claims10
A composition comprising phase separated nanoparticles comprising:
- liquid-ordered (Lo) domains comprising 1,2-distearoyl-sn-glycero-3-phosphocholine (DSPC);
- liquid-disordered (Ld) domains comprising 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC);
- a saturated PEG-modified lipid selected from 1.2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[methoxy (polyethylene glycol)-2000] (18:0 PEG2000-PE), 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[methoxy (polyethylene glycol)-1000] (18:0 PEG1000 PE), 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[methoxy (polyethylene glycol)-500] (18:0 PEG500 PE), and 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[methoxy (polyethylene glycol)-5000] (18:0 PEG5000 PE);
- cholesterol;
- and
- an unsaturated lipid conjugated to TNF-related apoptosis inducing ligand (TRAIL) at 1 mol %;
- wherein a ratio of the DSPC to the DOPC is 2:1 or 3:1.
