Abstract
An ex vivo method of determining sensitivity of multiple myeloma cells obtained from a multiple myeloma patient to chemotherapeutic agents, and methods of specifically treating the patient with chemotherapeutic agents to which the multiple myeloma cells are sensitive.
Claims17
A method for assessing sensitivity of multiple myeloma cells obtained from a subject to chemotherapeutic agents in an ex vivo assay, comprising:
- obtaining multiple myeloma cells from a subject as part of cellular components of a bone marrow aspirate from the subject;
- contacting the multiple myeloma cells obtained from the subject with at least one chemotherapeutic agent ex vivo for a period of 48 hours;
- incubating the multiple myeloma cells with the at least one chemotherapeutic agent in a serum free media comprising low-density lipoproteins to which interleukin-6 (IL-6) has been added, to form incubated cells;
- labeling at least one protein selected from the group consisting of CD138, CD38, CD45, and CD19, on the surface of the incubated cells to form labeled multiple myeloma (MM) cells;
- and
- analyzing the labeled MM cells by flow cytometry to determine the sensitivity and resistance of the subject's multiple myeloma cells to the at least one chemotherapeutic agent.
