Abstract
A composition including modified macrophages and its use in the treatment of melanoma and pancreatic cancer. The modified macrophages have a deletion of fragile-site associated tumor suppressor (FATS) or include a small interfering RNA (siRNA) capable of silencing the FATS. The knockout or inhibition of FATS may promote the polarization of M0 macrophages into M1 macrophages and suppress the polarization of M0 macrophages into M2 macrophages. The FATS can be used as a target for the production of medications for the treatment of cancer or tumors.
Claims16
A composition, comprising modified macrophages,
- wherein the modified macrophages have a deletion of fragile-site associated tumor suppressor (FATS) of SEQ ID NO:
- 1 or comprise a small interfering RNA (siRNA) capable of silencing the FATS of SEQ ID NO:
- 1,
- wherein the siRNA is SEQ ID NO:2.
A method for treating melanoma in a subject, comprising:
- a) introducing a small interfering RNA (siRNA) capable of silencing fragile-site associated tumor suppressor (FATS) of SEQ ID NO:
- 1 into macrophages to produce modified macrophages, wherein the siRNA is SEQ ID NO:2;
- and
- b) administering a therapeutically effective amount of a composition comprising the modified macrophages into the subject in need thereof, thereby treating the melanoma.
A method for treating pancreatic cancer in a subject, comprising:
- administering a therapeutically effective amount of a composition comprising modified macrophages into the subject in need thereof, thereby treating the pancreatic cancer,
- wherein the modified macrophages have a deletion of fragile-site associated tumor suppressor (FATS) of SEQ ID NO:
- 1.
