Abstract
The present disclosure provides methods, compositions, apparatuses and kits comprising ALDC enzymes having a better stability and/or activity, and, optionally, the yield of ALDC enzymes which can be recovered from microorganisms is improved. In some embodiments, the present disclosure provides methods, apparatuses, compositions and kits for the use of metal ions to increase stability and/or activity, and which further can be used to recover the enzymes from microorganisms in improved yields.
Claims2
A method of preparing an acetolactate decarboxylase (ALDC) enzyme preparation, the method comprising the steps of:
- culturing a Bacillus subtilis host cell comprising an expression cassette comprising a Bacillus subtilis aprE promoter and a Bacillus subtilis aprE signal peptide operably linked to a heterologous Brevibacillus brevis ALDC gene, in a cultivation medium supplemented with zinc at a concentration of 60 μM to 150 μM, whereby the host cell secretes the ALDC enzyme into the cultivation medium;
- and
- removing the host cell from the cultivation medium, thereby producing a cell-free ALDC enzyme preparation.
