Abstract
The present invention is directed to methods for determining the likelihood that source tissue will be suitable for the generation of viable precursor cells.
Claims18
A method of determining suitability of a source tissue for generating hair-follicle derived neural precursors (HFNs), the method comprising:
- obtaining a reference glucose consumption profile from intact human hair follicles that are capable of generating a viable HFN cell line;
- obtaining a sample comprising hair follicles from the source tissue;
- measuring a glucose consumption level of the hair follicles in the sample;
- comparing the measured glucose consumption level of the hair follicles in the sample to the reference glucose consumption profile;
- determining that:
- the source tissue is suitable for generating HFNs when the measure glucose consumption level is equal to or greater than that of the reference glucose consumption profile;
- or
- (ii) the source tissue is unsuitable for generating HFNs when the measured glucose consumption level is less than the reference glucose consumption profile, optionally obtaining viable hair-follicle derived neural precursors (HFNs) therefrom.

A method of determining if a sample of hair follicles comprises sufficient numbers of HFNs for isolation, the method comprising:
- obtaining a sample comprising hair follicles;
- measuring a glucose consumption level of the hair follicles in the sample;
- comparing the measured glucose consumption level of the hair follicles in the sample to a reference glucose consumption profile from intact human hair follicles from which HFNs were successfully isolated;
- determining that (i) the sample of hair follicles comprises sufficient numbers of HFNs for isolation when the measured glucose consumption level of the follicles in the sample is equal to or greater than that of the reference glucose consumption profile;
- or (ii) the sample of hair follicles does not comprise sufficient numbers of HFNs for isolation when the measured glucose consumption level of the follicles is less than that of the reference glucose consumption profile, optionally obtaining viable hair-follicle derived neural precursors (HFNs) therefrom.

An improved method of generating neuronal precursor cells from a sample of hair follicles or a sample of hair follicle cells, the method comprising:
- treating a sample of hair follicles or a sample of hair follicle cells in conditions enabling a transition of hair follicle cells to a growth phase, thereby forming a sample of conditioned hair follicles or hair follicle cells;
- measuring a glucose consumption level of the conditioned hair follicles or hair follicle cells;
- comparing the measured glucose consumption level of the conditioned hair follicles or hair follicle cells to a reference glucose consumption profile representative of hair follicles or hair follicle cells from which viable HFNs were isolated;
- and
- treating the conditioned hair follicles or hair follicle cells in conditions enabling enrichment of cells expressing neuronal lineage biomarkers when the measured glucose consumption level of the conditioned hair follicles or hair follicle cells is equal to or greater than that of the reference glucose consumption profile,
- thereby generating a composition of neuronal precursor cells from a sample of hair follicles or a sample of hair follicle cells.
