Abstract
Improvements in acoustically dispensed samples that are injected into an open port probe (OPP) are described. Apparatus and method are described that calibrate the volume dispensing determination and mechanisms in the acoustic dispenser to produce accurate and precise volumetric delivery.

A method of monitoring a reaction in situ, comprising:
- providing a reaction well containing a reaction mixture undergoing a reaction;
- dispensing a plurality of droplets from the reaction well at respective time points during the reaction without quenching the reaction;
- ionizing each dispensed droplet;
- and
- measuring, using a mass spectrometer, an amount of at least one reaction component in each ionized droplet.

A system for in situ kinetic monitoring of a chemical or biochemical reaction, the system comprising:
- a droplet dispenser operative to dispense a plurality of droplets from a reaction well at respective time points while a reaction proceeds in the reaction well;
- a capture probe for receiving and capturing each dispensed droplet with a transport fluid;
- an ionization source for ionizing each captured droplet;
- and
- a mass spectrometer for measuring an amount of at least one reaction component in each ionized droplet.
