Abstract
Described is a low voltage, pulsed electrical stimulation device for modulating the expression of sirtuin (“SIRT”), a useful protein, by tissues. Also described are methods of enhancing expression of SIRT in cells, particularly a method of stimulating the expression and/or release of SIRT in a cell having a gene encoding SIRT, wherein the method includes applying a bioelectric signal of, within 15%, 50 μA to about 500 μA, 20 pps, at 400 μsec pulse duration to the cell (e.g., directly, indirectly, or wirelessly). In order to downregulate expression of SIRT, the at least one bioelectric signal has a current of at least about 1 mA, as measured at the level of the living cells.
Claims16
A method of treating muscle atrophy in a subject, the method comprising:
- applying to skeletal muscle tissue of the subject a bioelectric signal produced by a bioelectric stimulator comprising an electric signal generator and at least one electrode,
- wherein the bioelectric signal:
- comprises a biphasic pulse,
- has a frequency within ±15% of 20 Hz,
- has a pulse width within ±15% of 400 microseconds (μsec),
- has a current of about 50 microamps (μA), and
- is administered for about one hour,
- thereby increasing expression of sirtuin 6 (SIRT6) in skeletal muscle cells and treating muscle atrophy in the subject.
A method of treating muscle atrophy in a subject, the method comprising:
- applying to skeletal muscle tissue of the subject a bioelectric signal produced by a bioelectric stimulator comprising an electric signal generator and at least one electrode,
- wherein the bioelectric signal:
- comprises a biphasic pulse,
- has a frequency within ±15% of 20 Hz,
- has a pulse width within ±15% of 400 μsec,
- has a current of from about 50 μA to about 500 μA, and
- is administered for from about thirty (30) minutes to about one hour,
- thereby increasing expression of SIRT6 in skeletal muscle cells and treating muscle atrophy in the subject.
A method of modulating expression of sirtuin 6 (SIRT6) in skeletal muscle cells of a subject, the method comprising:
- applying to skeletal muscle tissue of the subject a first bioelectric signal produced by a bioelectric stimulator comprising an electric signal generator and at least one electrode,
- wherein the first bioelectric signal:
- comprises a biphasic pulse,
- has a frequency within ±15% of 20 Hz,
- has a pulse width within ±15% of 400 μsec,
- has a current of less than 575 μA, and
- is administered for at least thirty (30) minutes,
- thereby increasing expression of SIRT6 in skeletal muscle cells,
- and wherein applying a second bioelectric signal having the same frequency and pulse width but having a current of at least 1 milliamp (mA) inhibits expression of SIRT6 in skeletal muscle cells.
