Abstract
A method for analyzing a cell surface glycan is provided, the method including bringing a glycan-binding substance labeled with a nucleic acid into contact with the cell and detecting the nucleic acid labeled to the glycan-binding substance bound to the cell, in which a kind and a quantity of the nucleic acid correspond to a kind and a quantity of the cell surface glycan.
Claims12
A method for analyzing at least one cell, the method comprising:
- preparing a mix of glycan-binding substances for analyzing glycans on a surface of subject cells, each glycan-binding substance of the mix of glycan-binding substances being labeled with a respective nucleic acid, each nucleic acid having a specific base sequence which specifies the glycan-binding substance, and corresponds to a kind of glycan, the mix including ten or more kinds of glycan-binding substances;
- bringing the mix into contact with the subject cells in a solution, whereby a bound portion of the mix binds to the glycans of the subject cells while an unbound portion of the mix remains in the solution;
- removing the unbound portion of the mix from the solution;
- isolating a single cell from the subject cells after the steps of bringing and removing, the subject cells and the single cell being alive during the step of isolating;
- detecting the nucleic acids labeled on glycan-binding substances that are associated with the single cell;
- and
- determining glycan profile of the single cell that is alive based on the detected nucleic acids and their quantities as indicators of kinds and quantities of the glycans on the surface of the single cell.

- contacting the mix with subject cells that are alive in a solution, whereby a portion of the mix binds to the glycans of the subject cells while a remaining portion of the mix remains unbound in the solution;
- washing away the remaining portion of the mix;
- isolating a single cell from the subject cells after the steps of contacting and washing;
- detecting nucleic acids labeled on glycan-binding substances that are associated with the single cell and RNA information in the single cell, wherein the detected nucleic acids and their quantities correspond to kinds and quantities of the glycans on the surface of the single cell that is alive;
- and
- obtaining a set of data of the kinds and quantities of the glycans and expressed genes of the RNA information for the single cell.
