Abstract
Provided is a pretreatment method for structural analysis of a protein, including chemical modifying and fragmenting a protein to be analyzed, in which in the fragmenting, the protein is treated at a temperature of 60 to 99° C. in the presence of a heat-resistant protease that has four or more cleavage sites and is soluble or immobilized on a carrier.
Claims5
A pretreatment method for structural analysis of a protein, the method comprising:
- a) chemically modifying a protein to be analyzed;
- and
- b) fragmenting the protein to be analyzed,
- wherein in step a), chemically modifying is a chemical modification of the protein by an electrophile,
- wherein in step b), the protein is treated at a temperature of 60° C. to 99° C. in the presence of a heat-resistant protease that has four or more cleavage sites and is soluble or immobilized on a carrier, and
- wherein the method further comprises removing one or more selected from the electrophile, a by-product of the electrophile, a quencher for the electrophile, and a by-product of the quencher for the electrophile, using an adsorbent.
