Abstract
Implants are described which comprise lipophilic pharmaceutical substances. Excipients are used which provide appropriate and controllable/tunable release of the lipophilic drug from the matrix of the implant. The implants can be implanted into a patient for release of the pharmaceutical substances. Methods of making and using such implants are also described.
Claims38
An implant comprising a non-biodegradable matrix, and a mixture of a lipophilic pharmaceutical substance and at least one excipient, the mixture dispersed in the matrix, wherein the matrix includes a non-biodegradable polymer, wherein the lipophilic pharmaceutical substance comprises a lipid conjugated to a pharmaceutical substance, wherein the at least one excipient includes a sugar alcohol, and wherein the implant is generally rod-shaped.
An implant comprising a non-biodegradable matrix, and a mixture of a lipophilic pharmaceutical substance and at least one excipient, the mixture dispersed in the matrix, wherein the matrix includes a non-biodegradable polymer, wherein the at least one excipient includes a sugar alcohol, wherein the lipophilic pharmaceutical substance, when present in saline, forms micelles of the lipophilic pharmaceutical substance, and wherein the implant is substantially generally rod-shaped.
An implant comprising a non-biodegradable matrix, and a mixture of an amphiphilic pharmaceutical substance and at least one excipient, the mixture dispersed in the matrix, wherein the matrix includes a non-biodegradable polymer, wherein the at least one excipient includes a sugar alcohol, and wherein the implant is generally rod-shaped.
A subcutaneous implant comprising:
- about 40% to about 60% by weight of a non-biodegradable matrix including ethylene vinyl acetate, and
- about 60% to about 40% by weight of a mixture of mannitol and a lipidated peptide comprising a glucagon-like peptide 1 analog,
- wherein said mixture comprises about 10% to about 30% by weight mannitol and about 90% to about 70% by weight of lipidated peptide, and wherein the implant is generally rod-shaped.
