Abstract
The present invention relates to a method of treating, preventing or delaying the progression of cancer and/or tumour in a subject comprising administering to the subject a treatment regimen comprising an effective amount of modified immunoresponsive cells expressing or presenting a heterologous T-cell receptor (TCR) having the property of binding to MAGE A4.
Claims21
A method of treating, preventing progression of, or delaying progression of synovial sarcoma in a subject, the method consisting of administering to the subject an effective amount of modified immunoresponsive cells presenting a heterologous T-cell receptor (TCR), the heterologous TCR comprising:
- (i) a TCR alpha chain variable domain comprising:
- a complementarity determining region 1 (αCDR1) comprising the amino acid sequence VSPFSN (SEQ ID NO:11);
- a complementarity determining region 2 (αCDR2) comprising the amino acid sequence LTFSEN (SEQ ID NO:12);
- and
- a complementarity determining region 3 (αCDR3) comprising the amino acid sequence CVVSGGTDSWGKLQF (SEQ ID NO:13);
- and
- (ii) a TCR beta chain variable domain comprising:
- a complementarity determining region 1 (βCDR1) comprising the amino acid sequence KGHDR (SEQ ID NO:14);
- a complementarity determining region 2 (βCDR2) comprising the amino acid sequence SFDVKD (SEQ ID NO:15);
- and
- a complementarity determining region 3 (βCDR3) comprising the amino acid sequence CATSGQGAYEEQFF (SEQ ID NO:16);
- wherein the synovial sarcoma expresses a peptide antigen of melanoma-associated antigen A4 (MAGE-A4) comprising the amino acid sequence GVYDGREHTV (SEQ ID NO:
- 2);
- and
- wherein the modified immunoresponsive cells do not express or present a heterologous CD8 co-receptor.
