Abstract
A method of reducing glycation of an amino acid in a protein produced by a host cell in cell culture medium is disclosed.
Claims18
A method of producing E25, an anti-IgE antibody, with reduced glycation comprising:
- (a) culturing Chinese Hamster Ovary (CHO) cells expressing the E25 antibody in a cell growth phase,
- (b) culturing the CHO cells in a production bioreactor in a fed batch culture with a cell culture medium comprising at least one reducing sugar at a concentration between 1.5 g/L and 3 g/L, and
- (c) harvesting the E25 antibody from the cell culture.
A method of producing an anti-CD20 antibody, with reduced glycation comprising:
- (a) culturing CHO cells expressing the anti-CD20 antibody in a cell growth phase,
- (b) thereafter culturing the CHO cells in a production bioreactor in a fed batch culture with a cell culture medium comprising at least one reducing sugar at a concentration between 1.5 g/L and 3 g/L, and
- (c) harvesting the anti-CD20 antibody from the cell culture.
