Abstract
A method is useful for diagnosing AIG, which includes the step of detecting whether autoantibodies against the beta-subunit of the gastric H+/K+-ATPase are present or not in a sample comprising antibodies. The method utilizes a diagnostically useful carrier that contains a solid phase on which an agent for the specific capture of the autoantibody is immobilized. Furthermore, the diagnostically useful carrier includes a solid phase on which an agent for the specific capture of an autoantibody against the beta-subunit of the gastric H+/K+-ATPase is immobilized.
Claims19
A method, comprising:
- incubating a sample comprising antibodies from a patient having or suspected of having autoimmune gastritis with a carrier comprising a solid phase, the carrier having immobilized thereon an agent for specific capture of an autoantibody against a beta-subunit of gastric H+/K+-ATPase, wherein said agent is only provided from or is a mammalian cell and is one or more selected from the group consisting of:
- an isolated or purified polypeptide comprising the beta-subunit of human gastric H+/K+-ATPase, or SEQ ID NO:
- 3 or SEQ ID NO:
- 5;
- and fixed cells comprising a polypeptide comprising the beta-subunit of human gastric H+/K+-ATPase, or SEQ ID NO:
- 3 or SEQ ID NO:
- 5;
- optionally wherein any of the polypeptides have a tag or detectable label, and
- detecting whether an autoantibody against the beta-subunit of gastric H+/K+-ATPase is present or not in the sample,
- wherein the beta-subunit of gastric H+/K+-ATPase is the sole biomarker tested for in the sample and the sole biomarker immobilized on the carrier.
A method, comprising:
- incubating a sample comprising antibodies from a patient having or suspected of having autoimmune gastritis with a carrier comprising a solid phase, the carrier having immobilized thereon an agent for specific capture of an autoantibody against a beta-subunit of gastric H+/K+-ATPase,
- wherein the agent is selected from the group consisting of:
- SEQ ID NO:
- 3 or SEQ ID NO:
- 5;
- and
- fixed cells comprising SEQ ID NO:
- 3 or SEQ ID NO:
- 5, and
- detecting whether an autoantibody against the beta-subunit of gastric H+/K+-ATPase is present or not in the sample,
- wherein the method does not comprise detecting whether an autoantibody against an alpha-subunit of gastric H+/K+-ATPase is present or not in the sample.
- 3;
- SEQ ID NO:
- 3 having a tag or detectable label;
- and
- fixed cells comprising SEQ ID NO:
- 3 or SEQ ID NO:
- 3 having a tag or detectable label, and
- detecting whether an autoantibody against the beta-subunit of gastric H+/K+-ATPase is present or not in the sample,
- wherein the method does not comprise detecting whether an autoantibody against an alpha-subunit of gastric H+/K+-ATPase is present or not in the sample.

- 5;
- SEQ ID NO:
- 5 having a tag or detectable label;
- and
- fixed cells comprising SEQ ID NO:
- 5 or SEQ ID NO:
- 5 having a tag or detectable label, and
- detecting whether an autoantibody against the beta-subunit of gastric H+/K+-ATPase is present or not in the sample,
