Abstract
The present invention relates to DNA polymerases. In particular, the present invention relates to heat labile DNA polymerases of marine origin, having high polymerase activity, strand displacement activity and 3-5′ exonuclease activity. Furthermore, the present invention provides heat labile DNA polymerases substantially without strand-displacement activity.
Claims8
An isolated DNA polymerase, wherein said DNA polymerase is the amino acid sequence selected from the group consisting of SEQ ID NOs:
- 3, 4, 5, 6, 7, and 8, or is at least 98% identical to the amino acid sequence selected from the group consisting of SEQ ID NOs:
- 3, 4, 5, 6, 7, and 8, wherein
- the amino acid in position 450 of SEQ ID NO:
- 3 is Asp,
- the amino acids in positions 449 and 451 of SEQ ID NO:
- 4 are Ala,
- the amino acids in positions 449 and 450 of SEQ ID NO:
- 5 are Ala and Asp, respectively,
- the amino acids in positions 450 and 451 of SEQ ID NO:
- 6 are Asp and Ala, respectively,
- the amino acids in positions 449, 450 and 451 of SEQ ID NO:
- 7 are Ala, Asp and Ala, respectively, and
- the amino acid in position 521 of SEQ ID NO:
- 8 is Ala
- wherein the DNA polymerase which is at least 98% identical to the amino acid sequence selected from the group consisting of SEQ ID NOs:
- 3 4, 5, 6, 7 and 8 only contains conservative amino acid substitutions;
- and
- wherein said DNA polymerase has no strand displacement activity.
