Abstract
The invention relates to methods for producing an RSV F protein trimer in a fed batch cell culture.
Claims25
A method for producing an RSV F protein trimer, comprising the steps of:
- (i) culturing mammalian cells that contain a gene encoding an RSV F protein in a cell culture medium, wherein said culturing comprises a growth phase and a production phase, and
- (ii) providing, during the growth phase and/or the production phase, glucose in a restricted manner to the cell culture by feeding glucose to the cell culture in response to rise of pH above a predetermined pH value in excess of a pH set point, wherein feeding glucose to the cell culture continues until the pH decreases to reach the pH set point of the culture, wherein the RSV F protein comprises an engineered disulfide mutation.
