Abstract
The disclosure relates to systems and methods for detecting and correcting errors in pH meter calibration or function when changing the pH of a sample, comprising measuring an initial pH, adding two or more amounts of titrant and measuring corresponding pH values, and applying a model to determine if the measured pH values correspond to pH values predicted by the model. The systems and methods can be used to control pH during viral inactivation of protein samples.
Claims33
A method comprising:
- measuring an initial pH (pHinitial) of a sample;
- adding a first amount of titrant (Titrantn) to the sample and measuring a first additional pH value (pHn), Titrantn being an amount of titrant added to the sample to reach pHn, wherein pHn is different from pHinitial, and wherein the initial pH (pHinitial) and the first additional pH value (pHn) are measured using a pH probe or a pH meter of a pH flow cell;
- applying a model to determine a second amount of titrant (Titrantn+1) to reach a target pH value, based on pHinitial, Titrantn and pHn, wherein the target pH is a first intermediate target pH;
- and based upon the result obtained by the model for determining a second amount of titrant (Titrantn+i), adding the second amount of titrant (Titrantn+i) to the sample;
- measuring a second pH (pHn+1) of the sample, using a pH probe or a pH meter of a pH flow cell, after adding the second amount of titrant (Titrantn+i);
- and
- correcting the pH meter-used to measure the second pH (pHn+1) when a difference between the second pH (pHn+1) and the target pH value to indicate a calibration error in the pH meter.
